Por um lado, pretende-se estudar e conhecer o alcance do equipamento avan\u00e7ado de digest\u00e3o din\u00e2mica in vitro relativamente \u00e0 sua capacidade de prever o perfil de solubilidade gastrointestinal de mol\u00e9culas incorporadas em matrizes complexas, tais como f\u00e1rmacos com formula\u00e7\u00f5es de liberta\u00e7\u00e3o controlada ou formula\u00e7\u00f5es do tipo pellets. Deste modo, pretende-se realizar estudos in vitro com o referido digestor sobre este tipo de formula\u00e7\u00f5es farmac\u00eauticas, para, assim, conhecer e validar a capacidade do digestor para prever o perfil de dissolu\u00e7\u00e3o desta tipologia de f\u00e1rmacos. Esta atividade n\u00e3o foi abordada no projeto anterior DIALFARMA, que se centrou em mol\u00e9culas de liberta\u00e7\u00e3o imediata.   <\/p>\n<p>Por outro lado, este projeto tem como objetivo desenvolver um dispositivo avan\u00e7ado no estudo da simula\u00e7\u00e3o da absor\u00e7\u00e3o intestinal, no qual seja contemplada a import\u00e2ncia da fluidodin\u00e2mica, tanto nos compartimentos luminais como nos basolaterais, sobre a absor\u00e7\u00e3o final dos compostos contidos nos digeridos intestinais. Assim, prop\u00f5e-se desenvolver um dispositivo espec\u00edfico que simule o fluxo dos conte\u00fados intestinais, aplicando um fluxo de entrada e retirada cont\u00ednua dos conte\u00fados gastrointestinais do l\u00famen intestinal. Da mesma forma, ser\u00e1 aplicado um fluxo na face basolateral, emulando assim o fluxo sangu\u00edneo da veia porta, que irriga o intestino, retirando os compostos que tenham sido absorvidos durante a digest\u00e3o.  <\/p>\n<p>Al\u00e9m disso, pretende-se realizar o estudo da viabilidade de um sistema integrado que contemple o funcionamento simult\u00e2neo do novo digestor din\u00e2mico in vitro avan\u00e7ado, j\u00e1 desenvolvido na primeira fase do projeto DIALFARMA, juntamente com o dispositivo de simula\u00e7\u00e3o da absor\u00e7\u00e3o intestinal. Gra\u00e7as a este sistema integrado, ser\u00e1 realizada a simula\u00e7\u00e3o do processo de digest\u00e3o gastrointestinal simultaneamente com a absor\u00e7\u00e3o intestinal, pelo que a digest\u00e3o intestinal do jejuno e do \u00edleo ser\u00e1 realizada sobre um conte\u00fado intestinal que ter\u00e1 experimentado progressivamente um processo de absor\u00e7\u00e3o intestinal. Este sistema representar\u00e1 um avan\u00e7o nos ensaios de digest\u00e3o gastrointestinal in vitro, ao contemplar simultaneamente a digest\u00e3o e a absor\u00e7\u00e3o, constituindo assim um aspeto diferenciador face \u00e0s tecnologias dispon\u00edveis no mercado, que geralmente realizam a absor\u00e7\u00e3o intestinal em condi\u00e7\u00f5es est\u00e1ticas ou a incorporam no processo de digest\u00e3o gastrointestinal in vitro atrav\u00e9s da utiliza\u00e7\u00e3o de membranas de di\u00e1lise ou de filtra\u00e7\u00e3o.  <\/p>\n<p>&nbsp;<\/p>\n<h2>Atividades<\/h2>\n<p>1. Avalia\u00e7\u00e3o do potencial de previs\u00e3o do equipamento de digest\u00e3o din\u00e2mica in vitro como ferramenta \u00fatil para a previs\u00e3o de formula\u00e7\u00f5es alimentares e\/ou farmac\u00eauticas complexas<br \/>\n2. Desenvolvimento e conce\u00e7\u00e3o de um dispositivo de simula\u00e7\u00e3o da absor\u00e7\u00e3o intestinal in vitro<br \/>\n3. Valida\u00e7\u00e3o do sistema integrado composto pelo digestor din\u00e2mico in vitro avan\u00e7ado e pelo dispositivo de absor\u00e7\u00e3o intestinal in vitro desenvolvido<br \/>\n4. Gest\u00e3o e coordena\u00e7\u00e3o do projeto<br \/>\n5 . Comunica\u00e7\u00e3o e divulga\u00e7\u00e3o de resultados<br \/>\n6 .
