Parmi celles-ci, les essais <strong data-start=\"549\" data-end=\"572\">in vitro et in vivo<\/strong> sont deux m\u00e9thodologies cl\u00e9s, chacune pr\u00e9sentant ses propres caract\u00e9ristiques, applications et avantages.<\/p>\n<h2 data-start=\"110\" data-end=\"177\"><strong>\u00c9tudes in vivo : d\u00e9finition et exemples d\u2019application<\/strong><\/h2>\n<p data-start=\"739\" data-end=\"951\"><strong>Que signifie in vivo ?<\/strong> Le terme in vivo vient du latin et signifie \u00ab dans le vivant \u00bb. Il d\u00e9signe les \u00e9tudes exp\u00e9rimentales r\u00e9alis\u00e9es sur des<strong> organismes vivants entier<\/strong>s, tels que des animaux ou des \u00eatres humains.<\/p>\n<p data-start=\"953\" data-end=\"1213\">Ces essais permettent d\u2019observer les effets d\u2019une intervention (comme un compos\u00e9, un m\u00e9dicament ou un ingr\u00e9dient fonctionnel) dans le contexte r\u00e9el d\u2019un syst\u00e8me biologique intact, o\u00f9 les interactions complexes entre cellules, tissus et organes sont conserv\u00e9es.<\/p>\n<p data-start=\"1215\" data-end=\"1290\">Selon l\u2019objectif de l\u2019\u00e9tude, <strong>diff\u00e9rents mod\u00e8les animaux sont s\u00e9lectionn\u00e9s :<\/strong><\/p>\n<ul data-start=\"1292\" data-end=\"1878\">\n<li data-start=\"1292\" data-end=\"1462\">\n<p data-start=\"1294\" data-end=\"1462\"><em data-start=\"1294\" data-end=\"1319\"><strong>Drosophila melanogaste<\/strong>r<\/em> (mouche du vinaigre) est largement utilis\u00e9e en recherche g\u00e9n\u00e9tique et neurocomportementale en raison de la facilit\u00e9 de manipulation g\u00e9n\u00e9tique.<\/p>\n<\/li>\n<li data-start=\"1463\" data-end=\"1630\">\n<p data-start=\"1465\" data-end=\"1630\"><strong><em data-start=\"1465\" data-end=\"1478\">Danio rerio<\/em><\/strong> (poisson-z\u00e8bre) permet d\u2019\u00e9tudier le d\u00e9veloppement embryonnaire, la toxicologie et la fonction g\u00e9n\u00e9tique gr\u00e2ce \u00e0 des techniques comme l\u2019\u00e9dition g\u00e9nique.<\/p>\n<\/li>\n<li data-start=\"1631\" data-end=\"1878\">\n<p data-start=\"1633\" data-end=\"1878\"><strong>Les rongeurs<\/strong> (souris et rats) et, pour des \u00e9tudes plus sp\u00e9cifiques, les primates non humains. Bien qu\u2019isol\u00e9s, ces mod\u00e8les conservent une partie de l\u2019architecture et de la fonctionnalit\u00e9 de leur environnement d\u2019origine, et sont utiles pour des \u00e9tudes physiologiques plus avanc\u00e9es que les mod\u00e8les classiques in vitro.<\/p>\n<h3 data-start=\"375\" data-end=\"449\"><strong>\u00c9tudes in vivo versus in vitro : utilisation des cultures cellulaires<\/strong><\/h3>\n<p data-start=\"4376\" data-end=\"4571\">Pour pr\u00e9server les caract\u00e9ristiques physiologiques, biochimiques et g\u00e9n\u00e9tiques du syst\u00e8me biologique d\u2019origine, les chercheurs ont souvent recours \u00e0 des<a href=\"https:\/\/www.ainia.com\/fr\/ainia-news\/types-de-culture-cellulaire\/\"> <strong>cultures cellulaires humaines ou animales.<\/strong><\/a><\/p>\n<p data-start=\"4573\" data-end=\"4697\">Ces cultures permettent de simuler des fonctions sp\u00e9cifiques de tissus ou d\u2019organes, et se classent en quatre grands ind\u00e9finiment.<\/p>\n<\/li>\n<li data-start=\"4885\" data-end=\"4951\">\n<p data-start=\"4887\" data-end=\"4951\"><strong>Hybridomes,<\/strong> utilis\u00e9s pour la production d\u2019anticorps \u00e0 l\u2019aide de cultures tridimensionnelles (3D), de bior\u00e9acteurs ou <a href=\"https:\/\/www.ainia.com\/fr\/ainia-news\/ingenierie-cellulaire-et-tissulaire-echafaudages\/\"><strong>d\u2019\u00e9chafaudages biofonctionnels,<\/strong><\/a> il est possible intestinale<\/strong> pour les ingr\u00e9dients, les nutraceutiques ou les additifs alimentaires.<\/p>\n<\/li>\n<li data-start=\"6099\" data-end=\"6288\">\n<p data-start=\"6101\" data-end=\"6288\"><strong>Tests de corrosion, d\u2019irritation et de p\u00e9n\u00e9tration<\/strong> cutan\u00e9e dans le d\u00e9veloppement de produits chimiques ou cosm\u00e9tiques, en utilisant vitro<\/strong> offrent de nombreux avantages qui en font des outils fondamentaux pour la<strong> recherche biom\u00e9dicale et pharmacologique.<\/strong><\/p>\n<ul>\n<li data-start=\"6950\" data-end=\"7245\">Elles permettent un <strong>contr\u00f4le exp\u00e9rimental plus rigoureux<\/strong>, r\u00e9duisant l\u2019interf\u00e9rence de variables syst\u00e9miques susceptibles d\u2019alt\u00e9rer l\u2019interpr\u00e9tation des r\u00e9sultats. Cela conduit \u00e0 une reproductibilit\u00e9 accrue par rapport aux mod\u00e8les in vivo, facilitant l\u2019obtention de donn\u00e9es coh\u00e9rentes et fiables.<\/li>\n<li data-start=\"7247\" data-end=\"7409\">En outre, l\u2019utilisation de tissus humains issus de chirurgie ou de dons conf\u00e8re une <strong>pertinence physiologique plus directe et repr\u00e9sentative<\/strong> de l\u2019organisme humain.<\/li>\n<li data-start=\"7411\" data-end=\"7580\">Autre avantage :<strong> l\u2019int\u00e9gration de techniques non invasives<\/strong> permettant l\u2019\u00e9tude dynamique de processus cellulaires et tissulaires sans compromettre la viabilit\u00e9 du mod\u00e8le.<\/li>\n<\/ul>\n<h3 data-start=\"545\" data-end=\"581\"><strong>Limites des \u00e9tudes in vitro<\/strong><\/h3>\n<p data-start=\"7615\" data-end=\"7995\">D\u2019un <strong>point de vue \u00e9thique<\/strong>, les <strong>\u00e9tudes in vitro<\/strong> sont une alternative valable et pr\u00e9f\u00e9rable \u00e0 l\u2019exp\u00e9rimentation animale, conform\u00e9ment au principe des <strong>3R (Replacement,\u00a0Reduction,\u00a0Refinement)<\/strong>. Cela encourage la r\u00e9duction du recours aux animaux en recherche en les rempla\u00e7ant par des m\u00e9thodes alternatives, en diminuant leur nombre et en am\u00e9liorant leurs conditions exp\u00e9rimentales.<\/p>\n<p data-start=\"7997\" data-end=\"8064\">Cependant, ces techniques pr\u00e9sentent aussi des <strong>limites importantes.<\/strong><\/p>\n<ul>\n<li data-start=\"8066\" data-end=\"8266\">L\u2019une des principales est<strong> l\u2019absence de r\u00e9ponse immunitaire ou syst\u00e9mique globale<\/strong>, ce qui limite la capacit\u00e9 \u00e0 reproduire certaines interactions biologiques complexes pr\u00e9sentes chez l\u2019organisme entier.<\/li>\n<li data-start=\"8268\" data-end=\"8486\">De plus, la manipulation de tissus humains n\u00e9cessite des <strong>conditions strictes<\/strong> de conservation et de manipulation pour maintenir leur viabilit\u00e9 et leur fonctionnalit\u00e9, ce qui implique des d\u00e9fis techniques et logistiques.<\/li>\n<\/ul>\n<h2 data-start=\"583\" data-end=\"652\"><strong>In vitro versus in vivo : approches compl\u00e9mentaires en recherche<\/strong><\/h2>\n<p data-start=\"8554\" data-end=\"8954\">Les mod\u00e8les <strong>in vivo, ex vivo et in vitro<\/strong> ne doivent pas \u00eatre consid\u00e9r\u00e9s comme des approches exclusives, mais comme des<strong> outils compl\u00e9mentaires<\/strong> dans l\u2019\u00e9cosyst\u00e8me de la recherche biom\u00e9dicale, technologique et appliqu\u00e9e. Chacun apporte des avantages sp\u00e9cifiques en fonction de l\u2019objectif de l\u2019\u00e9tude, du niveau de complexit\u00e9 requis, des exigences r\u00e9glementaires ou du type de compos\u00e9 ou produit \u00e0 \u00e9valuer.<\/p>\n<p data-start=\"8956\" data-end=\"9489\">Dans un contexte o\u00f9 la rigueur scientifique, l\u2019\u00e9thique en recherche et l\u2019efficacit\u00e9 dans le d\u00e9veloppement de solutions innovantes sont prioritaires,<strong> les essais in vitro gagnent en importance<\/strong> par rapport aux mod\u00e8les animaux.
